Origin

The Problem Came First

In 2022, Steven Watts was working on LNP formulation at a gene therapy contract research organization in the Minneapolis area, supporting programs targeting the liver — siRNA, mRNA, base editors. The formulations worked well. Then a client asked whether the same platform could be adapted for a Huntington's disease program that needed CNS delivery after intravenous dosing.

The answer, after six months of honest evaluation, was no. The ionizable lipid pKa ranges optimized for hepatocyte endosomal escape underperformed in cortical neuron assays. The ApoE-dependent uptake mechanism relied on fenestrated liver sinusoids — an architecture the BBB specifically lacks. The platform had been built for one organ, and the BBB was a different problem requiring different engineering from the start.

Biopathio was founded in Minneapolis in early 2024 to address that problem directly. The formulation approach starts with neural cell models, not hepatocytes. The surface engineering starts with BBB receptor biology — TfR1, LRP1, receptor-mediated transcytosis — not passive accumulation. The characterization panel includes BBB cell uptake and neuronal editing efficiency, not just DLS and RiboGreen.

Biopathio is a delivery platform company. We are not a drug developer. We do not develop gene therapies for specific CNS indications — we engineer the delivery vehicles that make CNS gene therapy possible for the teams that do.
